How much PROT1 should I use?

Ensure that the ratio of PROT1 to the blood sample is 1.4 to 1 and that the PROT1 is at room temperature when it is used.  As long as the ratio remains 1.4 to 1 this can be scaled up to larger volumes of blood:

  • 14 milliliters of PROT1 for 10 milliliters of whole blood (ie. when investigating extremely rare populations such as cancer stem cells)

or scaled down to smaller volumes of whole blood:

  • 140 microliters of PROT1 for 100 microliters of whole blood (ie. when banking samples from pediatric patients)

Is it OK to store in Falcon 50 mL polypropylene tubes?

Yes, use labware made of polypropylene or polyethylene such as a cryovial or conical tube.  If you are going to ship PROT1 you should probably look into the Nalgene "PASSPORT IP2" series like part number 2099-0004  https://www.usplastic.com/catalog/item.aspx?itemid=135938
